Ces dernières années, il a été
démontré que les axones sont
sensibles à des facteurs diffusibles
produits par des cellules situées à
distance de l'axone. Ces facteurs
peuvent être attractifs ou
répulsifs, parfois les deux. Des
travaux récents indiquent que la
nature répulsive ou attractive
d'une molécule sécrétée dépend
des récepteurs exprimés par le
cône de croissance mais aussi de
son état d'activation. La
concentration intracytoplasmique
en nucléotides cycliques et en
calcium semblent être les facteurs
essentiels contrôlant le guidage
axonal. Growing axons are guided by a variety of molecules which can either be diffusible or bound to a substrate or a membrane. Diffusible factors have been identified in several protein families, including the Netrins, Semaphorins, Neurotrophins, and Slit proteins, whose members can act as growth cone repellents or attractants, or, in some cases, both. Different classes of transmembrane receptors for these molecules have also been recently characterized. An important emerging question is to understand what specifies the positive or negative response of a growth cone to a given diffusible factor. Recent studies have shown that transmembrane receptors, and more specifically their cytoplasmic domains, dictate whether a growth cone sees a cue as attractive or repulsive. In addition, studies of the cytoplasmic signal transduction pathways mediating repulsion and attraction in a variety of neurons and species indicate that the response of a growth cone depends on its activation state, and that many guidance molecules converge to the same signaling pathways which can be modulated by cyclic nucleotides.
