Le VIH-sida doit se comprendre comme un défi dont on ne peut atténuer ni la violence, ni la gravité. On peut toutefois constater que cette pandémie a suscité et édifié de nouvelles formes de solidarités qui ont bouleversé nos repères en termes de soins et de santé publique. Ces quelques acquis des années sida éclairent autrement la figure de la personne malade et nous permettent de comprendre avec d’autres références la dimension politique de la santé publique. Les notions de respect, de solidarité et d’équité se comprennent dans le cadre d’un partenariat, et parfois même d’une alliance, thérapeutique. La relation de soin s’est située dans une perspective jusqu’alors inédite. Nos avancées révèlent davantage les injustices dans l’accès aux traitements et fixent l’horizon de nos responsabilités morales à l’égard de toute personne malade. La lutte contre le sida doit désormais se comprendre en termes de justice et de moyens investis dans le partage des acquis, tout particulièrement auprès des communautés les plus vulnérables. Les situations d’extrême précarité, les questions relatives aux conditions d’accès aux soins, à l’information, aux protocoles expérimentaux comme aux traitements et la qualité d’existence des personnes constituent autant d’aspects tangibles des défis qu’il nous faut considérer, aujourd’hui encore, en termes d’urgence vitale.; Violence and gravity cannot be ignored when considering HIV and AIDS challenges. However, coping with this pandemic disease caused us to build up and experience new ways of solidarity that have transformed our views of medical care and public health. The few achievements of these years throw a new light on the figure of the ill person. They brought new references allowing a different understanding of the political stakes of public health. Partnership or even therapeutic alliance are now a framework to the understanding of respect, solidarity and equity. New viewpoints exist on the health care relationship. Our achievements show at first injustice in the access of treatments at the international level. They make clear our moral responsibilities toward every person that require a treatment. This idea should be emphasized when applied on the most vulnerable communities. The most urgent challenges that remain today are extreme precariousness situations, treatment access conditions, information, and quality of life.

FAK (focal adhesion kinase) est une protéine cytoplasmique ubiquitaire retrouvée au sein du complexe d’adhérence. Son activation par les intégrines mais aussi par différents facteurs de croissance, cytokines ou hormones se traduit par l’autophosphorylation d’un résidu tyrosine (397) libérant un site de liaison pour la protéine Src. Cette liaison va permettre l’activation des protéines du complexe d’adhérence, aboutissant à la transmission de signaux régulateurs pour la migration, la survie et la prolifération cellulaires. Un dérèglement de ce système peut limiter ainsi plusieurs fonctions cellulaires, voire entraîner l’apoptose, et pourrait participer aux processus de cancérogenèse. Cet article présente les mécanismes par lesquels FAK contribue à la régulation cellulaire.; Focal adhesion kinase (FAK) is a cytoplasmic protein tyrosine kinase localized to regions called focal adhesions. Many stimuli can induce tyrosine phosphorylation and activation of FAK, including integrins and growth factors. The major site of autophosphorylation, tyrosine 397, is a docking site for the SH2 domains of Src family proteins. The other sites of phosphorylation are phosphorylated by Src kinases. Phosphorylated FAK binds proteins of focal adhesion and can activate them directly or indirectly by phosphorylation. These activated proteins forming the FAK complex facilitate the generation of downstream signals necessary to regulate cell functions, like motility, survival and proliferation. Dysregulation of FAK could participate in the development of cancer. This review will focus upon the mechanisms by which FAK transmits biochemical signals and elicits biological effects.

L’embryon du nématode Caenorhabditis elegans représente un système expérimental de choix pour disséquer les mécanismes qui orchestrent les processus de division cellulaire dans un organisme multicellulaire. En effet, un faisceau d’approches cellulaires, génétiques, moléculaires et génomiques permettent d’aborder chez cet organisme nombre de questions fondamentales relatives à la division cellulaire. Certaines des molécules caractérisées chez ce nématode pourraient jouer un rôle important dans d’autres organismes, y compris l’être humain. Dès lors, la recherche fondamentale menée sur l’embryon de C. elegans devrait avoir, à terme, un impact significatif sur le développement de nouveaux outils diagnostiques et thérapeutiques.; The mechanisms orchestrating spatial cell division control remain poorly understood. In animal cells, the position of the mitotic spindle dictates cleavage furrow placement, and thus plays a key role in governing spatial relationships between resulting daughter cells. The one-cell stage Caenorhabditis elegans embryo is an attractive model system to investigate the mechanisms underlying spindle positioning in metazoans. In this review, the experimental advantages of this model system for an in vivo dissection of cell division processes are first discussed. Next, three lines of experiments that were conducted to dissect the mechanisms governing spindle positioning in one-cell stage C. elegans embryos are summarized. First, localized laser micro-irradiations were utilized to identify the forces acting on spindle poles during anaphase. This work revealed that there is a precise imbalance of pulling forces acting on the two spindle poles, with the forces acting on the posterior spindle pole being in slight excess, thus explaining the asymmetric spindle position achieved by the end of anaphase. Second, an RNAi-based fonctional genomic screen was carried out to identify novel components required for generating these pulling forces. This uncovered that gpr-1/gpr-2, which encode GoLoco-containing proteins, as well as the previously identified Ga subunits goa- 1/gpa-16, are required for generation of pulling forces on the spindle poles. Third, the zyg-8 locus was identified by mutational analysis to play a distinct role during anaphase spindle positioning. zyg-8 was found to encode a protein related to human Doublecortin, which is affected in patients with neuronal migration disorders. Moreover, ZYG- 8 is a microtubule-associated protein that stabilizes microtubules against depolymerization. Together, these experimental approaches contribute to a better understanding of the mechanisms orchestrating spatial cell division control in metazoan organisms.
